Its reputation rests on this long lineage of traditional, cultural use rather than on any modern medical role.

Chemical Composition of Mulungu Bark

Mulungu bark holds a rich mix of plant compounds — flavonoids, triterpenes, and alkaloids — that together draw scientific attention. The most studied are the erythrina alkaloids, a group prized by researchers for their biological activity. Among them, erythravine and (+)-11α-hydroxy-erythravine stand out: these are the two compounds most examined for calming, anxiolytic-like activity in laboratory and animal work.1

Erythrina velutina, a close relative in the same genus, shares a similar chemical blueprint — a sign of the qualities Brazilian tradition has long attributed to the erythrina species. As modern analysis maps these compounds, it increasingly echoes what traditional knowledge held about the plant's calming character.

What Does the Science Say About Mulungu?

Scientific interest in mulungu has centred on how its erythrina alkaloids affect the nervous system. In rodent studies, both whole-bark extracts and the isolated alkaloids erythravine and (+)-11α-hydroxyerythravine have produced calming, anxiolytic-like effects in standard anxiety tests such as the elevated T-maze and the light–dark box.1,2 The clearest signals come from the isolated alkaloids and from the closely related species Erythrina velutina; notably, the same line of research found no antidepressant effect, which is a useful reminder of how specific these findings are.3 Related work also found the alkaloids protected rodents against chemically induced seizures.4

On mechanism, laboratory studies show these alkaloids act mainly as potent blockers of neuronal nicotinic acetylcholine receptors — part of the nervous system's signalling machinery.5 A 2020 study looked specifically for an effect on the GABA and glutamate systems and did not find a meaningful one, pointing to the nicotinic receptors rather than the GABA pathway as the main target.6

Human evidence is far more limited. Only a couple of small trials — both in patients before wisdom-tooth surgery — have tested mulungu in people, and their results conflict: a small crossover study reported a subjective sense of calm with no change in heart rate or blood pressure,7 while a larger, more tightly controlled randomised trial found mulungu no more effective than a placebo.8 In short, there is no robust clinical evidence that mulungu reduces anxiety in people.
